چکیده
A simple, rapid, and efficient live cell surface labeling method has been developed that uses a direct conjugation between Sortase A expressed transiently at the cell surface and Sortase A specific binding peptide.

Anchoring of surface proteins to the cell wall of Staphylococcus aureus. Sortase catalyzed in vitro transpeptidation reaction using LPXTG peptide and NH(2)-Gly(3) substrates.
Staphylococcus aureus sortase anchors surface proteins to the cell wall envelope by cleaving polypeptides at the LPXTG motif. Surface proteins are linked to the peptidoglycan by an amide bond between the C-terminal carboxyl and the amino group of the pentaglycine cross-bridge. We find that purified recombinant sortase hydrolyzed peptides bearing an LPXTG motif at the peptide bond between threon...
متن کاملAnchoring of Azospirillum brasilense to hydrophobic polystyrene and wheat roots
The anchoring (irreversible attachment) of Azospirillium brasilense Cd to hydrophobic polystyrene and to root surfaces was compared. Live A. brasilense Cd cells attached in significantly greater numbers to roots than to polystyrene, regardless of treatments made to the surfaces or to the bacterial cells. Triton X-100, Na2EDTA and several bacterial -inhibitory substances reduced bacterial attach...
متن کاملSortase A as a tool to functionalize surfaces.
A widely accepted approach to combat surface fouling is based on the prevention of biofoulants to attach to a surface by the functionalization with poly(ethylene glycol) (PEG). The goal of this study was to generate a proof of concept for the enzymatic coupling of PEG to a peptide precoated surface by using the enzyme Sortase A (SrtA). A hydrophobic polystyrene surface was primed with anchoring...
متن کاملThe molecular switch that activates the cell wall anchoring step of pilus assembly in gram-positive bacteria.
Cell surface pili in gram-positive bacteria orchestrate the colonization of host tissues, evasion of immunity, and the development of biofilms. Recent work revealed that pilus assembly is a biphasic process wherein pilus polymerization is catalyzed by a pilus-specific sortase followed by cell wall anchoring of the pilus that is promoted by the housekeeping sortase. Here, we present molecular ge...
متن کاملStructure and function of a Clostridium difficile sortase enzyme
Sortase enzymes are responsible for covalent anchoring of specific proteins to the peptidoglycan of the cell wall of gram-positive bacteria. In some gram-positive bacteria (e.g. Staphylococcus aureus), sortases have been found to be essential for pathogenesis and their inhibitors are under development as potential novel therapeutics. Here we provide the first report on the structural characteri...
